Branched Chain Amino Acid Aminotransferase
The kinetics of transamination reactions catalyzed by pig heart “branched chain amino acid” aminotransferase was investigated. The activation of aged preparations by 2mercaptoethanol, previously noted, was associated with increases in both the maximum velocity and enzyme-substrate affinities. This implies a protein conformational change. The sharp pH optimum observed with standard assay conditi...
متن کاملAn unnatural amino acid that mimics phosphotyrosine.
By replacing the phosphate group in pTyr with an isoxazole carboxylic acid, a novel unnatural amino acid has been successfully synthesized. Subsequently, its incorporation into a known PPI (protein-protein interaction) inhibitor of STAT3 protein (ISS 610) generated I2 which showed reasonable anti-STAT3 activity in both fluorescence polarization and cell-proliferation experiments.
متن کاملAspartate: 2-oxoglutarate aminotransferase from trichomonas vaginalis. Identity of aspartate aminotransferase and aromatic amino acid aminotransferase.
Aspartate: 2-oxoglutarate aminotransferase from the anaerobic protozoon Trichomonas vaginalis was purified to homogeneity and characterized. It is a dimeric protein of overall Mr approx. 100000. Only a single isoenzyme was found in T. vaginalis. The overall molecular and catalytic properties have features in common with both the vertebrate cytoplasmic and mitochondrial isoenzymes. The purified ...
متن کاملThe interaction of pyridoxamine 5-phosphate with aspartate aminotransferase.
The binding of pyridoxamine 5-phosphate to the enzyme aspartate aminotransferase from pig heart was investigated by fluorescence spectroscopy. The substantial decrease in fluorescence intensity at 390 rnp that follows the interaction of pyridoxamine 5-phosphate with the apoenzyme was used to determine the alhnity constant. The effect of pH on the stability of the apoenzyme-pyridoxamine 5-phosph...
متن کاملMolecular cloning, expression and characterization of pyridoxamine-pyruvate aminotransferase.
Pyridoxamine-pyruvate aminotransferase is a PLP (pyridoxal 5'-phosphate) (a coenzyme form of vitamin B6)-independent aminotransferase which catalyses a reversible transamination reaction between pyridoxamine and pyruvate to form pyridoxal and L-alanine. The gene encoding the enzyme has been identified, cloned and overexpressed for the first time. The mlr6806 gene on the chromosome of a symbioti...
